Differential protein analysis of Yiwei decoction on hypothalamus in precocious female rats

Abstract
    [Objective] To study the associated protein of Yiwei decoction on the regulation of hypothalamus in precocious female rats and to explore the mechanism of Yiwei decoction at the molecular level.[Methods] Twenty female rats aged 4~6 months were randomly divided into two groups:normal group and castration group. Ten female rats aged 10~12 months were used as the old group. After the administration, the hypothalamus tissue of three groups of rats was taken and iTRAQ was used to isolate, analyze and identify the differential proteins of rat hypothalamus using isotopic labeling relative and absolute quantitative techniques.[Results] The hypothalamus proteins in the rats of the three groups were compared and analyzed. According to the Venn diagram analysis of several groups of data, there were 17 differentially expressed proteins expressed in the precocious group and the castration group. Through Volcano Plot filtration, significant differences in protein (FC>1.5 or <2/3, P<0.05) were determined; seven types of senescence-related proteins and their related KEGG pathways were screened out. They were plasminogen, α-crystallin B chain, Hsp70 binding protein 1, serum transferrin, unconventional myosin-Id, myelin oligodendrocyte glycoprotein, and protein Fbxo44.[Conclusion] Yiwei decoction plays an important role in up-regulating or down-regulating these seven kinds of significantly different proteins, which lays a foundation for studying the effect of Yiwei decoction in delaying the aging of target proteins.
