Effect of Danggui Buxue decoction on the expression of PKC-ζ in the liver and skeletal muscle of diabetic rats

Abstract
    [Objective] To explore the effect of Danggui Buxue decoction on the protein kinase C-ζ expression in the liver and skeletal muscle cell of diabetic rats.[Methods] Fifty male SD rats were randomly divided into normal control group (NC group), Danggui Buxue decoction control group (DG group), type 2 diabetes mellitus group (T2DM group), type 2 diabetes mellitus + Danggui Buxue Decoction group (T2DM + DG group), type 2 diabetes + pioglitazone group (T2DM + BG group). Type 2 diabetes mellitus rat models were established by high fat diets and intraperitoneal injection with sreptozotocin (STZ). After 5 weeks, measuring fasting blood glucose (FPG) and fasting insulin (FINS) in blood and using immunohistochemical method to detect the content of PKC-ζ in the liver, skeletal muscle in rats seach group.[Results] Compared with NC group, the FPG and FINS of T2DM group increased significantly. Compared with T2DM group, the FPG and FINS of T2DM+DG group and T2DM+BG group decreased. The expression of PKC-ζ in the liver and skeletal muscle cell membrane of T2DM group was decreased by immunohistochemistry, while the expression of PKC-ζ in T2DM+DG group and T2DM + BG group was higher than that of T2DM group. Except that, there was no significant difference between T2DM+DG group and T2DM +BG group.[Conclusion] Danggui Buxue decoction can improve insulin resistance and reduce blood sugar, which may be related to promoting the expression of PKC-ζin the liver and skeletal muscle cells.
