Protective effect of Baogan decoction on chronic chemical liver injury induced by carbon tetra chloride in mice

Abstract
    [Objective] To study the protective effect of Baogan decoction on chronic chemicai liver injury of mice induced by carbon tetrachloride (CCl4).[Methods] A total of 84 Kunming mice were randomly divided into normal group, model group, hugan tablet group, baogan decoction high, middle and low dose group, with fourteen in each group. Except for the normal group, the other groups were injected with 0.5%CCl4 at the abdominal cavity on the first day, the third day, the 10th day, the 16th day, the 22th day, the 25th day and the 28th day in the experimental period. The 10th day of molding began to Intragastric administration. Normal group and model group were given intragastric administration of distilled water. Hugan tablet group, high dose of baogan decoction, middle dose of baogan decoctionand low dose of baogan decoction intragastric administration of drugs. After intragastric administration of 30 d, mice were weighed, killed, and liver tissues and serum samples were collected. Then test the content of the experimental indicators, and calculate the liver index and hepatic tissue pathological examination.[Results] Compared with the normal group, the ALT, AST and MDA in the model group increased significantly (P<0.05). Pathological changes in liver tissue of model group showed the arrangement of liver cells was disordered. Many large hepatic cells showed fatty degeneration and multiple foci of necrosis. Compared with the model group, the ALT, AST in the high, middle and low dose groups decreased(P<0.05), and the AST/ALT increased (P<0.05), and the MDA in the middle and low dose groups decreased (P<0.05). Liver tissue and morphological structure were significantly improved in each dose group.[Conclusion] Baogan decoction can alleviate the damage degree of CCl4 on liver cells, in which the hepatoprotective effect of the middle dose group is the best. The mechanism of action is related to the inhibition of lipid peroxidation after liver injury.
