Effect of modified Wendan Decoction on gastrointestinal motility in rat with depression

Abstract
    [Objective] To investigate the effect of modified Wendan Decoction on gastrointestinal motility in rats with depression.[Methods] Thirty-six SD rats were randomly divided into normal group,model group and modified Wendan Decoction group,with twelve rats in each group. The model was established by using the solitary and chronic unpredictable stress method. The gastric motility and intestinal propulsion rate were used to detect the intestinal motility of rats. The ultrastructure of gastrointestinal tissue was detected by transmission electron microscopy.[Results] After 21 days,compared with the blank group,the scores of model group,modified Wendan Decoction group were lower than those of the blank group,with statistical significance (P<0.01);28 days after administration,the scores of model group were lower than those of the normal group,with statistical significance(P<0.01);Compared with the model group,the score of modified Wendan Decoction group was higher than that of the model group,with statistical significance(P<0.05).There was no significant difference in activity and preference for sugar and water among the groups before modeling. After 21 days of chronic stress,compared with the blank group,the sugar-water preference rate of the other two groups decreased significantly (P<0.01);after 4 weeks of drug intervention,compared with the model group,the sugar-water preference of rats in modified Wendan Decoction group increased significantly (P<0.01).Compared with the blank group,the gastric emptying rate and small intestinal propulsion rate of the model group were significantly lower than those of the blank group (P<0.01);compared with the model group,the gastric emptying rate and small intestinal propulsion rate of the modified Wendan Decoction group were increased(P<0.05). The results of transmission electron microscopy showed that compared with the blank group,the membrane structure of the gastric main cells in the model group was incomplete,the nucleus was severely condensed,the nucleoplasm was shifted,some organelles in the cytoplasm was collapsed,and the mitochondria were unevenly arranged. The rough of endoplasmic reticulum was expanded,and the ultrastructural changes were more obvious under electron microscope;compared with the model group,the expression of gastric main cells in modified Wendan Decoction group and fluoxetine group was obviously relieved. Compared with the blank group,the disintegration state of colonic mucosal cells in the model group was severely damaged. Compared with the model group,the colonic tissue of the rats in the modified Wendan Decoction group was tightly connected and close to the normal group.[Conclusion] Modified Wendan Decoction can improve the depressive state of depression model rats,enhance gastrointestinal motility and improve the ultrastructure of gastrointestinal tissue in depressive rats,It provides experimental support for modified Wendan Decoction in the treatment of depression to improve gastrointestinal motility.
