Protective effects of artesunate against PC12 cells with oxidative injury through inducing the activities of antioxidase |

Abstract
|
[Objective] To investigate the protective effects of artesunate on PC12 cells with H2O2-induced oxidative damage and study its possible mechanisms.[Methods] CCK8 assay was used to assess the cell relative survival rates of PC12 cells induced by H2O2. Lactate dehydrogenase (LDH) assay was used to measure the protective effect of artesunate on PC12 cells injury induced by H2O2 as well. Malondialde (MDA),reactive oxygen (ROS),superoxide dismutase (SOD),CAT,GSH-PX in cells were detected by spectroscopy.[Results] Artesunate had no effect on relative survival rates of cultured PC12 cells under the tested concentrations. Compared with H2O2-induced group,the cell relative survival rates,the activities of SOD,CAT and GSH-PX were significantly increased in artesunate groups (P<0.05);while the levels of LDH,SOD and MDA were greatly decreased in artesunate groups compared with H2O2-induced group (P<0.05) with a dose-dependent manner.[Conclusion] Artesunate in a concentration range of 0.1~1.6 mmol/L can act as an antioxidant to prevent the cellular injury induced by H2O2 in cultured PC12 cells without affecting the cell survival rates. |
