Abstract

[Objective] To study the effect of Apigenin (APG) on anemia and disorders of electrolyte metabolism in chronic renal failure(CRF) rats.[Methods] 100 SD rats were randomly divided into normal control group, model group and APG low-dose[10 mg/(kg·d)],medium-dose[20 mg/(kg·d)],high-dose[40 mg/(kg·d)] groups,n=20;the CRF rat model was established by intragastric administration of adenine solution for 21 days, the drugs were given by intraperitoneal injection for 28 days, once a day, while the normal control group and model group were given saline simultaneously at the same time. The levels of serum urea nitrogen (BUN),serum creatinine (SCr) and uric acid (UA) were measured, the kidney index was calculated. The level of red blood cells (RBC),hemoglobin (HGB),hematocrit (HCT),serum iron (SI),total iron binding capacity (TIBC) and ferritin (FER) in whole blood were measured;the level of potassium (K+),sodium (Na+),calcium (Ca2+) and phosphorus (P3+) in serum were measured. The pathological changes of renal tissue was observed by HE staining.[Results] Compared with model group, the contents of BUN,SCr,UA in serum of APG medium-/high-dose groups were significantly reduced;renal index was significantly reduced;the levels of RBC,HGB,HCT,SI,TIBC,FER were significantly increased;the level of Ca2+ in serum was significantly increased and the levels of K+,Na+,P3+ were significantly decreased;all of the difference above were significant (P<0.05 or P<0.01). Pathological examination showed that APG treatment can effectively inhibit renal tissue lesions in CRF rats.[Conclusion] APG can improve the pharmacological effects of anemia and electrolyte disturbance in CRF rats, which may be one of the mechanisms that APG can play a protective role against renal failure. |
