Study on the protective effect of crocin on testicular tissue injury caused by diabetes mellitus

Abstract
    [Objective] To investigate the protective effects of crocin on testicular tissue injury caused by diabetes mellitus.[Methods] The 100 SD rats were randomized according to body weight divided into normal control group,model group,Crocin[25,50,100 mg/(kg·d)] groups,the drugs were given by gastric administration for 12 weeks. The diabetic rat models were made by intraperitoneal injecting STZ[30 mg/(kg·d)] for 3d continuously. The general condition of the rats in every group was observed;the FBG,FINS,GHb were detected. The body weight was detected and the testis tissue index was calculated,the content of testosterone in serum was determined;the histopathological changes of testis tissue was observed by HE staining,the ultrastructure of testis tissue was observed by electron microscope,the apoptosis of testis tissue apoptosis was observed by TUNEL.[Results] After treatment with crocin 50~100 mg/(kg·d) for 12 weeks,the FBG decreased significantly,FINS increased significantly,GHb decreased significantly in type 2 diabetic rat models (P<0.05 or P<0.01);the body weight,the testis tissue index and the content of testosterone were significantly increased (P<0.05 or P<0.01). 12 weeks later,the ultrastructural changes such as atrophy of seminiferous tubules,loss of spermatogenic cells and reduction of spermatogenic cells in testis tissue of diabetic rats,and ultrastructural changes such as nuclear condensation,chromatin mass accumulation,and reduction of mitochondria were significantly improved;the number of apoptotic cells in the testis decreased significantly,the Apoptosis Index (AI) decreased significantly (P<0.05 or P<0.01).[Conclusion] Crocin can effectively inhibit testicular tissue damage in diabetic rats,which mechanism may be related to its role in lowering blood sugar.
