Abstract
|
[Objective] To analyze the effect of Shenjin Yigu method on L-type calcium channel activity and intrachondrocyte calcium concentration,and to explore the mechanism of mechanical-biological transduction of Shenjin Yigu method in the prevention and treatment of knee osteoarthritis (KOA).[Methods] Fifty Japanese white rabbits were randomly divided into five groups. The KOA model induced by stress disorder was simulated by modified Hulth model. The observation group was treated with Shenjin Yigu method,while the control group was treated with sodium hyaluronate injection into articular cavity. Calcium ion concentration in primary chondrocytes of each group was measured by confocal laser microscopy,and current density of L-type calcium channel in chondrocytes of each group was measured by patch clamp technique.[Results] Calcium concentration in primary chondrocyte of observation group and control group was higher than that of model group,with statistical significance (P<0.05). Tthe current density of L-type calcium channel in observation group and control group was higher than that of model group,with statistical significance (P<0.05).[Conclusion] Shenjin Yigu method can regulate the activity of L-type calcium channel on the surface of chondrocyte,increase the concentration of calcium ion in chondrocyte,and then promote the synthesis and metabolism of chondrocyte. This mechanism may be one of the important reasons for the effect of massage manipulation or similar physiotherapy. |
