Abstract
    [Objective] To investigate the effects and pharmacological mechanisms of Yuanhu Zhitong Oral Liquid (YZOL) on primary dysmenorrhea and provide experimental basis for its clinical application.[Methods] The rat model of primary dysmenorrhea was induced by oxytocin. The writhing response was used as an indicator to investigate the effects of different doses of YZOL on dysmenorrhea rats. Plasma levels of estradiol (E2),progesterone (Prog),malondialdehyde (MDA),superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were measured in rats to explore the mechanisms of YZOL treatment. Moreover,the levels of prostaglandin F (PGF),prostaglandin E2 (PGE2),thromboxane B2 (TXB2),6-keto-prostaglandin F (6-keto-PGF),Ca2+ and nitric oxide (NO) in the uterus of rats were examined.[Results] YZOL could significantly reduce the writhing times (P<0.05) and writhing incidence of dysmenorrhea rats. Compared with the model group,significantly lower levels of MDA,PGF,Ca2+ (P<0.05) and markedly higher activity of 6-keto-PGF (P<0.05) were observed in the YZOL-treated group.[Conclusion] These results indicated that YZOL had a significant analgesic effect on the rat model of primary dysmenorrhea,and its mechanisms might be closely linked to the regulation of plasma MDA level and the levels of PGF,6-keto-PGF,Ca2+ in the uterine tissue.
