Effect of Qiyin Heye Decoction on glucose metabolism and NF-κB signaling pathway in skeletal muscle of rats with abnormal glucose tolerance

Abstract
    [Objective] To discuss the effects of Qiyin Heye Decoction on glucose metabolism and NF-κB signaling pathway in skeletal muscle of rats with abnormal glucose tolerance.[Methods] A rat model of impaired glucose tolerance was established by using high-fat and high-sugar diet. The model rats were randomly divided into model control group 1 group and different dose compatibility group 9 groups according to their weight,and a normal control group was set. In this study,orthogonal experiment was used to select three factors of astragalus,capillaris and lotus leaf for three-level experiment L9(34). Fasting blood glucose,fasting insulin and insulin resistance index were measured before and 6 and 12 weeks after intervention. The transcription levels of IκB-α and NF-κBp65 gene in muscle tissue of each group were detected by real-time fluorescent quantitative PCR,and pathological changes of skeletal muscle tissue were observed.[Results] Compared with the model control group,the insulin resistance index was significantly decreased (P<0.05) in group A,D,E,F,G,and H after 6 weeks of administration and in group D,F,G,H,and I after 12 weeks of administration. The insulin resistance index showed a significant difference with time (P<0.01). Rats in the model control group had skeletal muscle edema and inflammatory cell infiltration,but the expression level of NF-κBp65 was significantly decreased compared with the control group (P<0.05).[Conclusion] There was an inflammatory response in skeletal muscle tissue of IGT rats,but the NF-κB signaling pathway was not activated. Qiyin Heye Decoction can improve insulin resistance in IGT rat model and prevent the evolution of IGT to T2DM,but its mechanism needs further study.
