Regulation of acupuncture manipulation on cell-electrophysiology of BKCa channel in aorta vascular muscle in spontaneously hypertensive rat

Abstract
    [Objective] To observe the effect of acupuncture manipulation on cell-electrophysiology of calcium-activated potassium channel in aorta vascular muscle in spontaneously hypertensive rats.[Methods] Twenty-four aged 16 weeks spontaneously hypertensive rats (SHR) were randomly divided into the model group,the acupuncture manipulation group and the none acupuncture manipulation group. Eight WKY of the same age rats were used as the normal control group(WKY group). In the acupuncture manipulation group,Renying (ST9) was punctured with twisting manipulation of 120 times per minute for 1 minute. In the non-manipulation group,no manipulation was applied after inserting the needles. The needles were retained for 15 minutes in the two groups. The blood pressure was measured by non-invasive tail manometer,the current,opening time and opening probability of BKCa channel were measured by in-outward single-cell patch clamp technique.[Results] The systolic and diastolic blood pressures of model group,acupuncture manipulation group and non-manipulation group were significantly higher than those of WKY group (P<0.05). The blood pressure values of acupuncture manipulation group were significantly lower than those of model group at 1,2,3 and 4 weeks after treatment (P<0.05). Systolic blood pressure and diastolic blood pressure in the non-manipulation group were lower than that in the model group only after 1 week of treatment (P<0.05). The open probability and open time of BKCa channel in the model group were significantly lower than those of the normal control group (P<0.05),and the open probability of the acupuncture manipulation group was higher than that of the model group(P<0.05). There was no significant difference between non-manipulation group and model group (P>0.05).[Conclusion] The twisting manipulation of 120 times/min on ST9 can affect the cell-electrophysiology of calcium-activated potassium channel in aorta vascular muscle in spontaneously hypertensive rats. The antihypertensive effect may be related to regulation of the BKCa channel opening activities.
