Abstract
|
[Objective] To determine the correlation between SHR brain damage and RhoA and rock-1,and to explore the Rho pathway mechanism of acupuncture intervention in SHR brain protection.[Methods] SHR rats were randomly divided into model control group,taichong acupoint group,and WKY rats were set as blank control group. The acupuncture intervention in the taichong point group lasted for 4 weeks,and the other two groups were given the same degree and the same time quantum of grasping during the intervention period. The changes of systolic blood pressure,diastolic blood pressure and mean arterial pressure were observed. Morphological changes of brain tissue were observed by HE staining. The expression levels of RhoA and rock-1 were determined by Western blot.[Results] During the intervention period,the systolic,diastolic,and mean pressures of the normal control group were within the normal blood pressure range. Compared with the normal control group,the systolic blood pressure,diastolic blood pressure and mean blood pressure of the model control group were higher,and there was a tendency to increase with the increase of the weeks of conception. Compared with the model control group,the Taichong group can effectively reduce systolic blood pressure and has the trend of reducing diastolic blood pressure and average pressure. Compared with the normal control group,the model control group was disordered in cell tissue,the degree of cell necrosis and interstitial edema was aggravated,and the expression of RhoA and ROCK-1 was increased. Compared with the model control group,the taichong group had regular arrangement of brain cells,cell necrosis and interstitial edema disappeared and the expression of RhoA and ROCK-1 decreased.[Conclusion] Acupuncture taichong can effectively reduce SHR blood pressure and improve brain damage,and its mechanism may be related to the inhibition of RhoA/rock-1 signaling pathway expression. |
