Effect of Huoxue Wentong Formula on oxidative stress and apoptosis of cardiomyocyte induced by hypoxia

Abstract
    [Objective] To observe the protective effect of Huoxue Wentong Formula on hypoxia-induced injury of H9c2 myocardial cells.[Methods] H9c2 myocardial cells were stimulated by hypoxia and serum-free,and treated with serum containing Huoxue Wentong Formula and blank serum for 4 hours. Cell viability,Caspase 3 and Caspase 8 activities,SOD and CAT activities,MDA content were measured. The changes of ROS were observed by DHE staining,and the apoptotic cells was detected by Hoechst staining. The significant differences among the groups were compared.[Results] Compared with the control group,hypoxic group H9c2 myocardial cell viability,the activities of SOD and CAT decreased significantly (P<0.01),Caspase 3 and Caspase 8 activities,MDA content increased significantly (P<0.01),while the cumulative ROS content and the number of cell apoptosis were increased markedly (P<0.01). However,the serum containing Huoxue Wentong Formula could increase notablely cell viability,SOD and CAT activities (P<0.01),reduce MDA level,Caspase 3 and Caspase 8 activities significantly (P<0.01),and alleviate ROS accumulation,inhibit cardiomyocyte apoptosis (P<0.01).[Conclusion] Huoxue Wentong Formula protected myocardial cells from hypoxia injury to a certain extent by inhibiting apoptosis and alleviating oxidative stress.
