Abstract
|
[Objective] To investigate the chemical constituents of the leaves of Aglaia testicularis,and search for new anticancer leading compounds.[Methods] Silica gel,RP-18 reversed-phase silica gel column chromatography,and recrystallization were used for separation and purification. Structural identification was performed based on nuclear magnetic resonance,mass spectrometry,and other spectral data. Cytotoxicity was determined by MTT method.[Results] Eighteen compounds were isolated from the ethanol extract of the leaves of Aglaia testicularis,and identified as aglain C,aglaxiflorin D,10-O-acetylaglain C,rocaglaol,odorine,odorinol,(2R,3R)-2,3-di- (3,4-dimethoxybenzyl)-butyrolactone,secoisolariciresinol dimethyl ether,pinoresinol,richenone,richenol,ursolic acid,eichlerianic acid,shoreic acid,3β-hydroxy-5α,8α-epidioxyergosta-6,22-diene,stimast-5-en-3β,7α-diol,4β,10α-dihydroxy aromaden-drane and 3-hydroxy-4',6,8-trimethoxyflavon. The IC50 values of rocaglaol on AGZY 83-a (human lung cancer cell lines) and SMMC-7721 (human liver cancer cell lines) were 0.013 and 1.571 μg/mL.[Conclusion] Compounds 1,3-5,7,9-12,17,18 were isolated from this plant for the first time,compound 4 showed significant cytotoxic activity on both cell lines. |
