Effect and mechanism of Jingwanhong Ointment on scald of diabetic foot in rats

Abstract
    [Objective] To investigate the effect of Jingwanhong Ointment on scald of diabetic foot in rats and its mechanism.[Methods] The diabetic rat model was copied,and then diabetic foot scald model was established. The experiment was divided into four groups:sham operation group,model group,Jingwanhong Ointment group,recombinant human epidermal growth factor external solution (rhEGF) group. After 14 days of administration,the macromorphology,volume and Wagner grading results of rats' feet were recorded before and after administration. The content of superoxide dismutase (SOD),malondialdehyde (MDA),nitric oxide (NO) and the expression of platelet-derived growth factor (PDGF) and transforming growth factor-β (TGF-β) mRNA in foot tissue were measured after administration.[Results] After modeling,the toe volume,Wagner classification,SOD content,and PDGF mRNA expression in the model group were significantly different from those in the sham operation group (P<0.01). Compared with the model group,Jingwanhong Ointment group could significantly reduce the toe volume of diabetic rats (P<0.05) after 5 days of administration. After 14 days of administration,Jingwanhong Ointment group could significantly promote the repair of ulcer wound tissue and epidermis,significantly reduce Wagner classification level (P<0.01),significantly increase the content of SOD in foot tissue and up-regulate the expression of PDGF mRNA (P<0.05).[Conclusion] Jingwanhong Ointment has multiple effects on controlling diabetic foot scald in rats,controlling infection,removing necrotic tissue,and promoting tissue repair. Its healing effect may be related to eliminating excess oxygen free radicals and up-regulating PDGF mRNA expression.
