Abstract
|
[Objective] Preliminarily analyze the chemical constituents of Wenxin Granules by ultra-high performance liquid chromatography-tandem quadrupole time of flight mass spectrometry (UPLC/Q-TOF-MS).[Methods] An ACQUITY UPLC BEH C18 column (2.1 mm×100 mm,1.7 μm) was used,with acetonitrile-water (containing 0.1% formic acid) as mobile phase gradient elution,flow rate 0.3 mL/min,electrospray ionization source (ESI source),positive/negative ion (Positive/Negative) dual mode detection. The molecular weights of compounds obtained by UPLC/Q-TOF MS/MS and MS/MS collision dissociation fragments were used to analyze and identify the main chemical components of Wenxin Granules combined with literature report.[Results] Combined with literature reports and primary MS and secondary MS/MS high resolution mass spectrometry data,a preliminary identification of 68 of these compounds was performed. The results showed that the main components of Wenxin Granules were sugars,saponins,flavonoids,among which,notoginsenoside R1,ginsenoside Rg1,Rb1 are abundant.[Conclusion] UPLC/Q-TOF MS/MS is an effective method to identify the components of traditional Chinese medicine compound. This study is the first time to use this technology to carry out qualitative analysis on the main components of Wenxin Granules,which provides an effective scientific basis for clarifying the material basis of the efficacy of compound Chinese medicine,exploring its mechanism of action and quality control. |
