Effect of Xian'ou Rupu Recipe on skin ulcer wound healing and mitochondria in diabetic rats

Abstract
    [Objective] To investigate the effect of Xian'ou Rupu Recipe on skin ulcer wound healing and mitochondria in diabetic rats.[Methods] Forty-eight male SD rats were made into a diabetic rat ulcer model by intragastric administration of high-fat emulsion,injection of streptozotocin (STZ),and production of full-thickness skin ulcers on the back. There were randomly divided into the model control group and the Xian'ou Rupu Recipe group. The Xian'ou Rupu Recipe group was coated with the powder of Xian'ou Rupu Recipe,while the model control group were not treated. On the 3rd,7th and 14th day after modeling,the rats were sacrificed and collected. The ultrastructure of mitochondria in wound fibroblasts was observed by transmission electron microscope,and the contents of adenosine triphosphate (ATP) and reactive oxygen species (ROS) in wound were measured by enzyme-linked immunosorbent assay (ELISA) and statistically analyzed.[Results] The wound healing rate of the Xian'ou Rupu Recipe group on the 7th and 14th days was higher than that of the model control group (P<0.05). The ultrastructure showed that the swelling degree of the Xian'ou Rupu Recipe group on the 3rd day was lighter than that of the model control group,and a few ridges disappeared. On the 7th day,the mitochondrial swelling of the Xian'ou Rupu Recipe group was slightly reduced,and the mitochondria of the clear ridge structure were more visible. On the 14th day,mitochondria were abundant and had clear structure. But on the 7th day,mitochondrial structure of model control group was broken more seriously,accompanied by aggregation,deformation,fusion and other changes. On the 14th day,the number of swollen mitochondria decreased,and some of them showed clear ridge structure. The results of ELISA showed that ATP was higher and ROS was lower in Xian'ou Rupu Recipe group than that in model control group on the 7th and 14th day (P<0.05).[Conclusion] Xian'ou Rupu Recipe can protect against mitochondrial damage,promote mitochondrial production of ATP and inhibit excessive production of ROS,improve mitochondrial energy supply,reduce the level of oxidative stress in wounds,and promote wound healing in diabetic ulcer rats.
