| [Objective] To investigate the effect of different doses of Lvfan Decoction on anxiety behavior after ischemic stroke in rats.[Methods] SPF male healthy SD adult rats were used the rat anxiety model of middle cerebral artery focal cerebral ischemia combined with empty bottle stimulation. The successfully prepared model rats were randomly divided into model group,buspirone hydrochloride group (3 mg/kg),Lvfan Decoction low (1.6 g/kg) and high-dose group (3.2 g/kg),and blank control group. Continuous intragastric administration for 14 days,and 1 hour after the last administration,observe the intervention effect of different doses of Lvfan Decoction on anxiety behavior of rats through behavioral experiments such as elevated cross maze,open field experiment,light and dark box test and Morris water maze.[Results] The elevated cross maze experiment showed that the time ratio and frequency ratio of rats entering the open arm in the model group were reduced,and the difference was significant statistically compared with the blank control group (P<0.01). Lvfan Decoction can obviously increase the total distance of rats in open field and the time of entering the central area,and has a tendency to increase the time ratio and frequency ratio of rats entering the open arm. Behavioral experiments,such as open field experiment,bright black box test and Morris water maze,all suggest that the compound model rats have anxious behavioral phenomena such as decreased autonomous activity and nervous fear. After the intervention of Lvfan Decoction,the autonomous activity and exploratory behavior of rats have increased,which is consistent with the previous clinical studies.[Conclusion] Lvfan Decoction has certain anti-anxiety effects,and the effect of high-dose Lvfan Decoction is more remarkable. |
