Effects of Siegesbeckia orientalis extract on the expression of AKT and ERK1/2 in myocardium of rats with coronary artery ligation

Abstract
    [Objective] Through the effect of Siegesbeckia orientalis extract on the expression of myocardial AKT and ERK1/2 in myocardial ischemic rat model,to explore its protective effect on myocardial ischemic injury in rats.[Methods] The 90 male SD rats were randomly divided into 6 groups,each with 15 rats,namely sham operation group,model control group,Siegesbeckia orientalis extract low-dose group,Siegesbeckia orientalis extract middle-dose group,Siegesbeckia orientalis extract high-dose group,isosorbide dinitrate group. The rat model of myocardial ischemia injury was constructed and ligated with 2 mm below the root of the left atrial appendage. After 4 weeks of continuous administration,blood was collected from the abdominal aorta of the rats to detect myocardial enzymes. The level of myocardial damage in the ischemic area was observed by HE staining. The expression of AKT and ERK1/2 in myocardial cells was detected by immunohistochemistry.[Results] Compared with the model control group,serum AST of Siegesbeckia orientalis extract low-dose group,middle-dose group and high-dose group were significantly reduced. Serum LDH of Siegesbeckia orientalis extract low-dose group,middle-dose group and high-dose group reduced,but no significant difference. Serum CK significantly decreased in the high-dose Siegesbeckia orientalis extract group;Serum CK-MB in the Siegesbeckia orientalis extract middle-dose group and high-dose group significantly decreased. The HE staining grade of myocardial tissue in the ischemic area of Siegesbeckia orientalis extracts in the low-dose,middle-dose and high-dose groups was significantly reduced. The expression of AKT in myocardial cells in the ischemic area of Siegesbeckia orientalis extract increased significantly in the low-dose,middle-dose and high-dose groups;the expression of ERK1 in myocardial cells in the ischemic area of Siegesbeckia orientalis extract increased significantly in the middle-dose group and high-dose group.[Conclusion] Siegesbeckia orientalis extract can increase the expression of AKT and ERK1 in the ischemic area of myocardium to produce a protective effect on myocardial injury.
