Effect of electroacupuncture at Renying and Taichong on kidney tissue Foxp3 and RORγt in spontaneous hypertension rats model

Abstract
    [Objective] To observe the effect of "Huoxue Sanfeng" acupuncture on the protein and mRNA expression of FoxP3 and RORγt in kidney tissue of prehypertensive rats. [Method] Sixteen 4-week-old spontaneously hypertensive rats were randomly divided into a model group (SHR group) and an electro-acupuncture group (EA+SHR group),8 rats in each group. 8 WKY rats served as a blank control group. In EA+SHR group,acupuncture were performed at Renying and Taichong bilaterally,once a day for 2 weeks. The Softron non-invasive blood pressure measurement was used to measure the systolic blood pressure before intervention,1 week and 2 weeks after EA treatment. HE staining was used to detect the morphology of rats renal. Western blot and PCR technology were applied to assessed the FoxP3,RORγt protein and mRNA expression. [Result] The level of systolic blood pressure in SHR rats was significantly higher than that in WKY group (P<0.05). After two weeks of intervention,the level of systolic blood pressure in EA+SHR group was significantly lower than that of SHR group (P<0.05). The expression of RORγt mRNA in kidney tissue of SHR group was significantly higher than that of WKY group (P<0.05),FoxP3 protein and mRNA expression,as well as the FoxP3/RORγt ratio was significantly lower than that of WKY group (P<0.05). The renal tissue RORγt mRNA expression of EA+SHR group was significantly lower than that of SHR group (P<0.05),FoxP3 protein expression and FoxP3/RORγt ratio were significantly higher than that of SHR group (P<0.05). There was no obvious change of renal morphology after EA intervention. [Conclusion] "Huoxue Sanfeng" acupuncture can regulate the expression of FoxP3 and RORγt,and the antihypertensive effect of EA on prehypertensive rats may be related to regulating the balance of FoxP3 and RORγt.
