Abstract
|
[Objective] The research on the isolation and characterization of polysaccharide from Schisandra chinensis is expected to provide scientific data for the further study of Schisandra chinensis.[Methods] The crude polysaccharides of Schisandra chinensis were obtained by water extraction and ethanol precipitation. A polysaccharide from Schisandra chinensis was further isolated and purified by graded ethanol precipitation and Sephadex G-150. Phenol-sulfuric acid method was used to determine the content of polysaccharide. High performance size exclusion chromatography coupled with multi-angle laser light scattering detector and refractive index detector method (HPSEC-MALLS-RID),high performance liquid chromatography (HPLC),infrared spectroscopy (IR) and nuclear magnetic resonance (NMR)were used to determine its molecular weight,monosaccharide compositions,functional groups and structure,respectively.[Results] A polysaccharide from Schisandra chinensis (SCP-3-1) was isolated from Schisandra chinensis. SCP-3-1 was mainly composed of mannose,galacturonic acid,glucose,galactose and arabinose in molar ratios of 0.09:0.19:19.82:2.08:0.13 with polysaccharide content of 74.28% and molecular weight of 84.9 kDa. IR and NMR analyses indicated that it was a pyran polysaccharide connected with α-glycosidic bonds.[Conclusion] This study can provide useful reference for further analyzing structure of polysaccharide,and also provide a scientific basis for the application of Schisandra chinensis. |
