Effects of Mangan Yangyin Capsule on the expression of Caspase-3 and PTP1B in liver tissues of alcoholic hepatitis rats and correlation study |

Abstract
|
[Objective] To observe the effect of Mangan Yangyin Capsules on rat alcoholic hepatitis model.[Methods] The 50 wistar rats were randomly divided into blank control group,model control group,and Mangan Yangyin Capsules(0.27 g/kg low-dose group,0.54 g/kg high-dose group). Polyene phosphatidylcholine capsule positive drug control group(0.15 mg/kg),except the blank control group,the other groups were intragastrically administered with 50% ethanol at 12 mL/kg. Each administration group was intragastrically administered with corresponding drugs,once a day,10 mL/kg,the blank control group and model control group were intragastrically administered with equal volume of distilled water. The drug was administered for 16 weeks while building the model. Take venous blood and test serum AST and γ-GT. After homogenizing 1g of liver tissue,extract the supernatant to detect the content of SOD and MDA in liver tissue. The remaining liver tissue was used for HE staining,and the expression of Caspase-3 and PTP1B in the liver tissue was detected by immunohistochemistry.[Results] Mangan Yangyin Capsules can significantly reduce the activity of AST and γ-GT in serum (P<0.05 or P<0.01). Mangan Yangyin Capsules can significantly increase the SOD activity in liver tissues (P<0.05 or P<0.01). Mangan Yangyin Capsules can significantly reduce the content of MDA in liver tissue (P<0.05 or P<0.01). Mangan Yangyin Capsules can significantly reduce the expression of Caspase-3 and PTP1B in liver tissues (P<0.05 or P<0.01). There were a certain number of fatty vacuoles in the liver tissues of Mangan Yangyin capsules in each treatment group,but fatty vacuoles and inflammatory cell infiltration were reduced to a certain extent compared with the model control group.[Conclusion] Mangan Yangyin Capsules may protect liver injury caused by heavy drinking by improving the body's antioxidant capacity,reducing insulin resistance,and anti-apoptosis. |
