Abstract
    [Objective] To investigate the expression and significance of tankyrases (TNKS) in Xiaoyan Decoction against lung adenocarcinomacells.[Methods] The expression of TNKS in lung adenocarcinoma tissues and adjacent tissues was detected by immunohistochemistry in 37 cases. qRT-PCR was used to detectthe mRNA expression of TNKS in A549 cells treated with gradient concentration of Xiaoyan Decoction. CCK-8 assay,γ-H2AX immunofluorescenceand DNA repair assay were used to detect the cell proliferation,DNA damage and DNA repair respectively in each group of A549 cells. The scratch test and transwell chambers were used to detect the migration and invasion capacity of cells in each group of A549 cells.[Results] 1) The positive expression rates of TNKS protein inlung adenocarcinoma tissues and paracancerous tissues were 63.00% and 18.92%,respectively.The differences were statistically significant (P<0.05). 2)The mRNA expression of TNKS in A549 cells decreased with the increase of Xiaoyan Decoction concentration.The differences were statistically significant(P<0.05). 3) After TNKS was down-regulated,the cell proliferative activity was decreased and the expression of γ-H2AX was increased.The ability of DNA repair,cell migration and invasion were all inhibited. Similarly,after cells were treated with Xiaoyan Decoction,the results were consistent with those in the TNKS down-regulated group.[Conclusion] TNKS are closely related to cellproliferation,DNA damage and repair,migration and invasion of lung adenocarcinomacells. The antitumor effect of Xiaoyan Decoction may be related to the down-regulation of TNKS expression.
