Investigation on the mechanism of salvianolic acid for injection in the treatment of ischemic stroke based on metabolomics

Abstract
    [Objective] To explore the mechanism of salvianolic acid for injection(SAI) in the treatment of ischemic stroke by metabolomics method. [Methods] UHPL-Q-Orbitrap MS was utilized to fulfill metabolomics in 39 patients' blood samples with ischemic stroke who met the corresponding inclusion and exclusion criteria in convalescent period,the plasma of patients before and after treatment of SAI was analyzed by unsupervised principal component analysis(PCA),and supervised orthogonal partial least squares discriminant analysis(OPLS-DA). [Results] A total of 42 biomarkers related to the treatment of ischemic stroke were screened out by metabolomics analysis,which involved 22 metabolic pathways. The significantly changed six metabolic pathways mainly included phospholipid biosynthesis,phosphatidylcholine biosynthesis,porphyrin metabolism,histidine metabolism,steroidogenesis,and pyrimidine metabolism. [Conclusion] Confirmed by metabolomic approach,the mechanism of SAI on the treatment of ischemic stroke was possibly related to the regulation of phospholipid metabolism,amino acid metabolism,nucleic acid metabolism and energy metabolism. And it is indicated that metabonomics is feasible to guide the clinical and mechanism research of traditional Chinese medicine.
