Abstract
    [Objective] To establish a method for simultaneous determination the content of syringin,costunolide and dehydrocostus lactone in vladimiriae radix by microwave extraction with UPLC analysis. [Methods] The best microwave extraction process of vladimiriae radix was optimized by taking the extraction rates of syringin,costunolide and dehydrocostus lactone as indexes,and twenty-two batches of vladimiriae radix were determined content by UPLC. [Results] The optimum microwave extraction conditions of vladimiriae radix as follows:methanol as solvent,microwave power 400 W,extraction time 2 min,and the liquid/solid ratio of 80:1 (mL/g). The extraction rates of three target compounds in different batches of vladimiriae radix under these conditions were higher than those by ultrasonic extraction. The method used to determine simultaneously the content of syringin,costunolide and dehydrocostus lactone in vladimiriae radix by UPLC showed good linearity,the RSD of the precision,repeatability and stability tests were less than 3% and the average recoveries were ranged in 101.79%~102.23%. The results showed that the content of three compounds in vladimiriae radix of different batches were different. It showed that there were large differences in the quality of vladimiriae radix on the market. [Conclusion] The method established in this experiment was stable,rapid and efficient with good repeatability,which could be used for the quality evaluation of vladimiriae radix.
