Banxia Xiexin Decoction inhibited ulcerative colitis in mice by inhibiting inflammation

Abstract
    [Objective] To observe the effects of Banxia Xiexin Decoction dose and modern clinical dose on inflammatory factors in mice with ulcerative colitis induced by dextran sulfate sodium (DSS-UC) free drinking. To explore the reasonable clinical dosage and immune mechanism of ulcerative colitis.[Methods] The 32 C57BL6/J mice were randomly divided into control group (n=8),model group (n=8),Banxia Xiexin Decoction ancient prescription group (n=8) and Banxia Xiexin Decoction pharmacopoeia prescription group (n=8). Ulcerative colitis model was induced by 2.5% DSS free drinking for 7 days. At the same time,the mice in each group were given normal saline,normal saline,Banxia Xiexin Decoction Gufang and Banxia Xiexin Decoction pharmacopoeia by using gastric gavage for 14 days. Histopathology was observed with hematoxylin-eosin staining (HE) and enzyme linked immunosorbent assay (ELISA) was then used to detect the levels of inflammatory factors in serum and colon tissues.[Results] Banxia Xiexin Decoction could significantly restore body weight and decrease disease activity index (DAI) index of DSS-UC model mice. The levels of 5-hydroxytryptamine (5-HT) and interleukin-10 (IL-10) in serum and colon tissue of DSS-UC model mice were increased,while interleukin-4 (IL-4) and tumor necrosis factor (TNF-α) were decreased,but interferon-gamma (IFN-γ) level was not significantly effect.[Conclusion] Banxia Xiexin Decoction could improve intestinal inflammatory response and tissue damage of DSS-UC mice by increasing the levels of IL-10 and 5-HT in serum and colon tissue,and reducing the levels of IL-4 and TNF-α in serum and colon tissue,which plays a therapeutic role in treating UC. Furthermore,the effect of Gufang dose is better.
