Protective effect of Shengmai Capsule on chemotherapy-induced intestinal mucositis in mice treated with 5-Fluorouracil

Abstract
    [Objective] To investigate the protective effect of Shengmai Capsule on colon mucosal damage induced by 5-fluorouracil(5-FU) in mice. [Methods] Twenty male Balb/c mice were randomly divided into normal group,model group and Shengmai group. The mice in model group and Shengmai group received intraperitoneal injection of 5-FU(25 mg/kg) to induce colon mucosal damage. Then,mice in normal group and the model group were given an equal amount of distilled water by gavage,while mice in Shengmai group was given an equal amount of Shengmai capsule suspension(350 mg/kg) by gavage every day. The body weight and diarrhea of mice were monitored every day;the colon length of mice was measured;the activity of diamine oxidase(DAO) in mice serum was detected;histopathological changes of colon were observed by HE(Hematoxylin-eosin staining) staining;the expression level of ki67 in colon tissue was detected by immunohistochemistry;The number of goblet cells in colon tissue was detected by PAS(Periodic Acid Schiff stain) staining. [Results] Compared with the model group,the diarrhea of mice in Shengmai group was improved significantly;the length of colon increased significantly(P<0.05);The activity of DAO in serum decreased(P<0.01);the structure of colon mucosa was improved obviously;the expression of ki67 and the number of goblet cells in colon tissue increased(P<0.000 1). [Conclusion] Shengmai capsule has obviously protective effect on colon mucosal damage induced by 5-FU.
