Abstract
    [Objective] To study the mechanism of Huangqi Guizi Wuwu Decoction(HGW) on alleviating the pathological changes caused by oligoasthenospermia,based on the modification of gut microbiota. [Methods] Normal Kunming mice were selected as control group,which received an 7-days injection of saline;the other mice were injected by 60 mg/(kg·d) I.p Cyclophosphamide continuously for seven days,which were divided into oligoasthenospermia model group and HGW low,middle and high dose treatment group(8,16,32 g/kg). Each group contained ten mice. H&E staining was used to observe the pathological changes in testis. The gut microbiota was detected by 16S rDNA sequencing. The body weight and testis quality would be recorded after 35 days intragastric administration. The sperm quality would be tested by computer-aided sperm analysis(CASA) software. [Results] AThe results showed that HGW greatly enhanced testis and epididymis index,significantly increased sperm quality and viability,reduced pathological lesions of testicular tissue in mice with oligoasthenospermia. Furthermore,they could also reverse the gut microbiota disorder,increase the relative abundance of Parabacteroides,decrease the relative abundance of Roseburia. [Conclusion] These results suggested that HGW could mprove the spermatogenic function and gut microbiota dysbiosis in mice with oligoasthenospermia induced by cyclophosphamide.
