| Effects of Xingnao Kaiqiao acupuncture on behavioral changes and release of monoamine neurotransmitters in post-stroke depression model rats |

| Abstract
|
| [Objective] To investigate the effects of Xingnao Kaiqiao acupuncture on behavioral changes and release of monoamine neurotransmitters in post-stroke depressed(PSD)rats. [Methods] A total of 56 SD rats were randomly divided into a normal group(14 rats), a model group(14 rats),a drug group(14 rats),and an acupuncture group(14 rats). Except for the normal group,the remaining 42 rats were used to prepare the PSD model using the Longa line occlusion method and the chronic unpredictable mild stress(CUMS)method, and rats that failed to model and died during the modeling process were excluded. The drug group was given fluoxetine intragastric treatment. The acupuncture group was treated by Xingnao Kaiqiao acupuncture,and the normal group and model group were given 1 mL normal saline intragastric treatment. Neurologic deficit score,body weight,vertical score and horizontal score in open field test,sugar water consumption and serum monoamine neurotransmitter release level were compared before modeling(0 d),before CUMS(7 d), before treatment(14 d)and after treatment(35 d). [Results] According to the ANOVA results of repeated measurement data,there were statistically significant differences in neurological impairment scores at different time points in the group(P<0.001),and there was an interaction between time and group(P<0.01). The results indicated that the variation trend of neural function deficit score in different groups was different with time. There was a statistical difference in body weight at different time points in the group(P<0.001),and there was an interaction between time and group(P<0.01),suggesting that the change trend of body weight in different groups was different with time. Compared at different,differences between the vertical scores and horizontal scores in open field experiment(P<0.001),and there was an interaction between time and group(P<0.01). The results indicated that the vertical and horizontal scores of rats in different groups were different over time. Compared at different time points in the group,there were statistical differences in sugar water consumption(P<0.001). There was an interaction between time and group(P<0.01),suggesting that the change trend of sugar water consumption in different groups was different with time. After treatment(35 d),the levels of 5-Hydroxytryptamine(5-HT),Dopamine(DA)and Norepinephrine(NE)in the model were significantly decreased(P<0.05),and the levels of 5-HT,DA and NE in the drug group and acupuncture group were significantly increased compared with the model group(P<0.05). [Conclusion] Acupuncture for awakening the brain and opening the mind is comparable to the effect of fluoxetine in improving behavioral and neurotransmitter aspects. The method of acupuncture has a certain recovery effect on nerve function injury. |
