Jianpi Huayu Decoction inhibits EMT in hepatoma cells via regulating the CD44/ERK pathway

Abstract
    [Objective] To study the inhibiting effect of Jianpi Huayu Decoction(JHD)in epithelial-mesenchymal transition(EMT)of hepatoma carcinoma cell,and to explore the possible mechanism behind it. [Methods] Culture MHCC97H cells in vitro,and divide the MHCC97H cells into three groups according to the stimulations(control group,10% of the drug-containing serum group,20% of the drug-containing serum group).The cell migration and invasion abilities was evaluated by scratch test and Transwell invasion test, and Real-time PCR and Western Blot were used to detect the expression levels of genes and proteins related to the EMT pathway. [Results]Compared with control group,the migration and invasion ability of the drug-containing serum groups were significantly decreased. As well as,the mRNA and protein expression level of Cluster of differentiation 44(CD44),Extracellular regulated kinase (ERK),zinc finger protein transcription factor(Snail),Vimentin and N-cadherin were significantly decreased,while the expression of E-cadherin was increased. [Conclusion] JHD may inhibits EMT,migration,and invasion of hepatocellular carcinoma MHCC97H cells through inhibiting the activation of the CD44/ERK pathway.
