| Abstract
|
| [Objective] Heart failure(HF)is the terminal stage of cardiovascular diseases,with high morbidity and mortality. The theory of“similar diseases and syndromes of damp-turbidity-phlegm-rheum”proposed by renowned master of traditional Chinese medicine(TCM)ZHANG Boli,provides valuable insights into the pathogenesis and treatment of HF. According to this theory,there is a potential correlation between HF risk and phlegm-dampness constitution. This study aims to explore plasma lipid biomarkers associated with heart failure risk in individuals with phlegm-dampness constitution and reveal their potential metabolic mechanisms using modern omics technologies. [Methods] This study is based on genome-wide association study(GWAS)data from international public databases. Mendelian randomization(MR)analysis was employed,combined with lipidomics data,to investigate the causal relationship between phlegm-dampness constitution and HF. Two-step MR analysis was further conducted to calculate the mediating effects of plasma lipid biomarkers between phlegm-dampness constitution and HF risk. [Results] The results show that phlegm-dampness constitution significantly increases the risk of HF(OR=1.205,P<0.001). Furthermore,56 plasma lipid metabolites were significantly associated with HF risk,of which 30 lipid biomarkers play a mediating role between phlegm-dampness constitution and HF risk. [Conclusion] Within the framework of the“similar diseases and syndromes of damp-turbidity-phlegm-rheum”theory,this study applied genetic causal inference to evaluate the association between metabolic traits related to phlegm-dampness constitution and heart failure risk,and identified plasma lipid biomarkers with potential mediating roles. These results provide methodological support for the quantitative study of constitution-related cardiovascular risk and offer data reference for constitution-based cardiovascular risk assessment research. |
