Exploration on the effective substances and mechanism of action of Yinhuo Tang in the treatment of lung adenocarcinoma based on UPLC-Q-TOF-MS,network pharmacology and experimental verification

Abstract
    [Objective] To explore the pharmacodynamic material basis and mechanism of Yinhuo Tang in treating lung adenocarcinoma. [Methods] The blood-entry components of Yinhuo Tang were analyzed by UPLC-Q-TOF-MS technology,and the component targets were obtained by combining PubChem and other databases. The targets of lung adenocarcinoma were obtained by using GeneCards, OMIM and other databases. The protein-protein interaction(PPI)network of potential targets for Yinhuo Tang in treating lung adenocarcinoma was constructed,and GO and KEGG enrichment analysis were carried out. Yinhuo Tang-containing serum was prepared to intervene in A549 and H1299 lung adenocarcinoma cell lines. CCK8 method was used to determine the optimal intervention concentration and time,and RAS activator ML-098 was used for rescue experiments. The cell migration ability was observed by scratch assay,and the protein expression of key molecules in MAPK/ERK pathway was detected by Western blot. [Results] The 21 blood-entry components of Yinhuo Tang were detected,and 574 potential drug targets,1 341 potential targets for lung adenocarcinoma treatment and 176 intersection targets were obtained. KEGG analysis showed that Yinhuo Tang might exert anti-lung cancer effects by affecting cancer,Ras,MAPK and other signaling pathways. In vitro experiments showed that Yinhuo Tang could inhibit the migration of A549 and H1299 lung cancer cells(P<0.05). Compared with the blank control group,the expression of key proteins in MAPK/ERK pathway in the Yinhuo Tang group was decreased(P<0.05). There was no significant difference between the Yinhuo Tang + ML-098 group and the blank control group(P>0.05),indicating that the inhibitory effect of Yinhuo Tang could be reversed by ML-098. [Conclusion] Yinhuo Tang can effectively inhibit the migration ability of lung adenocarcinoma cells,and its mechanism may be related to the MAPK/ERK signaling pathway.
