Exploring the effect of external ironing of silkworm sand on diabetic gastroparesis rats based on PI3K/AKT signaling pathway

Abstract
    [Objective] To explore the mechanism of the action of external ironing method on diabetic gastroparesis(DGP) rats. [Methods] The 8 rats were randomly selected as the blank group,and the rest of the rats were constructed as the DGP rat model,after successful modelling,they were randomly divided into the model group,the external ironing group(the external ironing method of the silkworm sand) and the mosapride group(1.57 mg/kg) and were treated continuously for 2 weeks. Detection of gastric emptying rate,random blood glucose level in rats,HE staining was used to observe the morphological changes in the gastric antrum,Western blot was used to detect the protein expression of phosphatidylinositol-3-kinase(PI3K),phosphorylated(p)-PI3K,protein kinase B(AKT),p-AKT,and TUNEL staining was used to detect the apoptosis of the gastric antrum in rats. [Results] Compared with the blank group,the gastric emptying rate of the model group decreased significantly(P<0.01);random blood glucose level increased(P<0.01);the glandular structure of the gastric sinus department was disturbed;p-PI3K/PI3K,p-AKT/AKT,protein expression increased significantly(P<0.01);apoptosis of the gastric sinus department was increased(P<0.01). Compared with the model group,gastric emptying rate of the externally ironed group increased significantly(P<0.01);random blood glucose decreased(P<0.05);the tissue structure of the gastric sinus department was gradually restored;the protein expression of p-PI3K/PI3K,p-AKT/AKT decreased significantly(P<0.01);the apoptosis of the gastric sinus department decreased(P<0.01). [Conclusion] External ironing of silkworm sand reduces apoptosis,improves gastric motility,and lowers random blood glucose levels in the gastric sinus of rats with diabetic gastroparesis,the mechanism of which may be related to the regulation of the PI3K/AKT signalling pathway.
