针刺对快速老化小鼠SAMP10氧化应激相关基因表达的影响

摘要:
[目的]探讨快速老化小鼠SAMP10脑衰老的机制,揭示“益气调血、扶本培元”针法改善SAMP10脑衰老症状的部分机制。[方法]应用cDNA阵列技术 (cDNAArray),观察快速老化小鼠SAMP10与正常老化小鼠SAMR1前脑588个基因表达的差异,同时观察“益气调血、扶本培元”针法对上述基因表达的影响。[结果]SAMP10与SAMR1相比,谷胱甘肽S转移酶、热休克蛋白86、成纤维细胞生长因子8、NF -kappa -B转录因子p65亚单位4个与氧化应激反应有关的基因表达上调,而针刺可使这些基因的表达下调。[结论]SAMP10的快速老化可能与其脑内的氧化应激水平升高有关,针刺可改善氧化应激水平进而达到延缓脑衰老的作用。

Influence of acupuncture on expression of relative oxidation stress gene in accelerated aged rat SAMP10

Abstract:
[Objective] To explore the partial mechanism of brain senescence in SAMP10,and the improvement function of“Yiqitiaoxue,fubenpeiyuan”on senile symptoms of brain.[Method] cDNA Array technique was used,and the expression difference of 588 genes of front brain was observed in SAMP10and SAMR1,meanwhile the influence of“Yiqitiaoxue,fubenpeiyuan”on the expression of the above mentioned gene was observed.[Results] Compared with SAMR1,the expression of glutathione transferase S,HSP86,fibroblast growth factor FGF,and NF-kappa-B p 65 subunit 4 gene relative to oxidation stress response in SAMP10was upregulated,while acupuncture could decrease the expression.[Conclusion] The rapid senescence of SAMP10may be related to the upregulated level of oxidation stress gene in the brain,acupuncture can improve it and so as to retard brain senescence.
