[目的]通过观察中药咳喘方对哮喘豚鼠肺组织嗜酸细胞凋亡的影响,进一步探讨中药咳喘方治疗哮喘的疗效机制。[方法]采用经典的哮喘豚鼠模型,将60只符合实验标准的雄性豚鼠随机分为4组,即正常对照组、模型组、泼尼松组和咳喘方组,镜下观察各组支气管肺泡灌洗液嗜酸细胞计数的变化及肺组织嗜酸细胞浸润和凋亡的情况,并采用免疫组化的方法观察各组肺组织嗜酸细胞Fas和bcl-2凋亡基因的表达情况,将结果进行统计学分析。[结果]咳喘方组支气管肺泡灌洗液嗜酸细胞计数和肺组织嗜酸细胞浸润的数量明显低于模型组和泼尼松组,而肺组织嗜酸细胞凋亡的数量则高于模型组和泼尼松组;咳喘方组肺组织嗜酸细胞凋亡基因Fas的表达较其他组增强,而凋亡基因bcl-2的表达较泼尼松组和模型组低。[结论]中药咳喘方能减少哮喘豚鼠肺组织嗜酸细胞的浸润,促进嗜酸细胞凋亡基因Fas的表达,诱导嗜酸细胞凋亡,这可能是其治疗哮喘的疗效机制之一。 |

Abstract: |
[Objective] To investigate the effect of the“Kechuan Fang”on eosinophils apoptosis of lung tissue in guinea-pig with asthma.[Method] Guinea pigs were randomly divided into four groups,asthma model group,prednisone group,“Kechuang Fang”group and normal control group.Eosinophils were isolated from the bronchovesicular lavage.Eosinophils and eosinophils apoptosis counts in every group were measured with biochemistry and immunohistochemistry.Fas-mRNA and bcl2-mRNA expression were determined by means of in situ blotting.The results were analyzed using colour- pathological image analysis system.[Results] The eosinophils counts and infiltration in“Kechuang Fang”group were significantly lower than those in both asthma model group and prednisone group,but eosinophils apoptosis counts were higher significantly than those in that two groups.Eosinophils Fas-mRNA expression of lung in the asthma guinea-pig in“Kechuang Fang”group increased more significant than that of others,but eosinophils bcl2-mRNA expression decreased more significant than that of both the asthma model group and prednisone group.[Conclusion] The“Kechuang Fang”can reduce eosinophils infiltration in asthma guinea-pig lung tissue,promote eosinophils apoptosis-gene Fas expression,and induce eosinophils apoptosis. |
