调肝导浊中药对体外培养大鼠主动脉平滑肌细胞增殖调控的影响

摘要:

[目的]观察调肝导浊中药对大鼠主动脉平滑肌细胞 (SMC)内Ca²⁺浓度、C_myc表达和SMC增殖的影响。[方法]采用体外SMC技术,免疫组织化学方法测量SMC的增殖核抗原(PCNA)阳性细胞率和C_myc阳性表达率,通过负荷钙离子荧光指示剂Fura₂/AM进行[Ca²⁺]i 测定。[结果]高脂血清培养组SMC的PCNA阳性细胞率和C_myc阳性表达率明显高于正常培养(P<0.01)。调肝导浊中药通过降低胞内[Ca²⁺]i,抑制C_myc表达,使该组的PCNA阳性率明显低于高脂血清组(P<0.01)。[结论]调肝导浊中药对SMC增殖起到良好的调控作用,从而有效地防止动脉粥样硬化 (AS)的发生与发展。

Abstract:

[Objective] To observe the influence of“Tiaogandaozhuo”herbs on concentration of Ca²⁺,expression of C_myc and proliferation of SMC.[Methods] PCNA positive cell rate and C_myc positive expression rate were detected by external SMC technique,and immunohistochemical method,[Ca²⁺] was determined by load fluorescence indicator Fura-2/AM.[Result] The PCNA positive cell rate and C_myc positive expression rate in high-fat serum culture group was higher than that of normal group(P<0.01).“Tiaogandaozhuo”herbs made the PCNA positive rate obviously lower than that of control group by descending [Ca²⁺]and inhibiting of the expression of C_myc,(P<0.01).[Conclusion] “Tiaogandaozhuo”herbs possess good regulating and controlling effect on the proliferation of SMC,and hence effectively prevent the occurring and development of atherosclerosis(AS).
