摘要:

[目的]观察全蝎提取液对急性早幼粒细胞株(HL-60)细胞及相关基因的影响。[方法]全蝎诱导HL-60细胞凋亡的形态学观察及流式细胞仪分析;全蝎对HL-60细胞周期分布的影响;免疫组化法观察全蝎对HL-60细胞凋亡调控基因表达的影响。[结果]全蝎可以明显促进HL-60细胞的程序化死亡,并在一定范围内呈正比关系,最高浓度(10m/mL)时,凋亡细胞比例明显增加,达21.44%.中、小剂量药物浓度时,细胞凋亡比例略有增加,分别为12.56%和9.47%,对照组仅7.36%.同时可显著提高P₅₃基因的表达水平(与对照组相比,P<0.01)和降低bcl-2基因的表达。[结论]全蝎提取液对HL-60细胞凋亡及相关基因的表达具有益影响。

Abstract:

[Objective] To observe the effect of Scorpio extract on progranulocyte(HL-60) in acute promyelocytic leukemia and the correlative gene. [Methods] Scorpio induced morphologic changes of apoptosis of HL-60 and flow cytometry analysis, the effect on the distribution in cellular cycle of HL-60, the effect on the expression of modulating genes in apoptosis of HL-60 determined with immuohistochemistry were investigated. [Results] Scorpio could markedly accelerate the programming death of HL-60. The positive dose effect correlation could be found in certain concentration range. At the highest concentration(10 mg/mL) the proportion of apoptosis increased obviously, peaking at 21.44%. When middle or small dosage were administered, the proportion increased slightly. They were 12.56% and 9.47% respectively. It was only 7.36% in control. Simultaneously, it could markedly increase the expression of P₅₃ gene(P<0.01 compared with control),and decrease the expression of bel 2. [Conclusion] Scorpio extract has beneficial effect on HL-60 death and correlative gene expression.
