[目的]观察针刺对多发梗死性痴呆(MID)模型大鼠大脑皮层bcl-2和baxmRNA及蛋白表达的影响。[方法]通过向颈内动脉注入自体血栓的方法建立大鼠MID模型。将大鼠随机分为模型组、针刺组、非穴组和假手术组。针刺组取人中、内关、风池穴,非穴组取双侧胁下两固定非穴点,针刺1次d/。模型组与假手术组进行与针刺组和非穴组相同时间、相同程度的捉抓。连续针刺2周后断头处死,取左侧大脑。原位杂交和免疫组化法观察皮层bcl-2和baxmRNA及蛋白的表达。[结果]模型组bcl-2和baxmRNA和蛋白的表达均增加;针刺后,bcl-2mRNA和蛋白的表达进一步增加,baxmRNA和蛋白的表达减少。[结论]针刺可促进MID模型大鼠大脑皮层bcl-2的表达,抑制bax的表达,从而保护脑组织,且这种作用具有穴位特异性。 |

Abstract: |
[Objective] To observe the influence of acupuncture on the expression of bax mRNA and protein of cerebral cortex in rats with multi.infarct dementia MID. [Methods] The MID in rats was established by infusion of autogeneic thrombus into carotid internal artery.The selected rats were randomly divided into four groups: model group,acupuncture group,non.acupoint group and sham operation group.In the acupuncture group Renzhong point,Neiguan point and Fengchi point were selected,while in the non.acupoint group two non.acupoint in the bilateral latus were used.The acupuncture was pricked once a day.The rats in the model and sham operation group were only caught and grasped at a same time interval and degree.Two weeks after treatment,the rats were killed by head.dislocation and the tissue of left brain was harvested.The expression of bcl.2,bax mRNA and protein was observed by in situ hybridization and immunohistochemistry. [Result] The expression of bcl.2,bax mRNA and protein in cerebral cortex in model group increased obviously.Further increase of bcl.2 mRNA and protein expression was found after acupuncture.But the expression of bax mRNA and protein decreased. [Conclusion] The acupuncture may promote the expression of bcl.2 mRNA and protein in cerebral cortex and inhibit the expression of bax mRNA and protein in cerebral cortex in rats with MID.This action may protect brain tissue and possesses acupoint specificity. |
