黄芩苷对脑缺血大鼠的治疗作用及对Caspase-3的影响

摘要:
[目的]通过TTC染色,动物神经行为学评分以及脑组织超微组织观察判断黄芩苷对局灶缺血性大鼠的治疗作用及对半胱氨酸蛋白酶-3(Caspase-3)的影响。[方法]用线栓法建立大鼠脑缺血再灌注模型,通过TTC染色,神经功能检查法对脑缺血后动物进行行为测评,病理及超微组织观察作为药效评价辅助手段,应用原为杂交技术检测Caspase-3的表达变化,准确判断黄芩苷对局灶缺血性大鼠的治疗作用及其作用机制。[结果]黄芩苷可以减少脑缺血后脑梗死面积,减轻脑水肿,减少脑组织海马、皮层Caspase-3的表达。[结论]黄芩苷具有神经脑保护作用,调节Caspase-3表达可能是使其发挥作用的重要机制之一。

Effect on Therapy and Expression of Caspase-3 of Baicalin in Rats with Cerebral Ischemia

Abstract:
[Objective] To estimate the therapeutic effect and the influence on the Caspase-3 of baicalin in rats with focal cerebral ischemia by TTC staining, through the evaluating the score of animal neural behavior and observing the cerebral tissue ultrastructure.[Methods] Ischemia and reperfusion in rats were established with thread occlusion. The score of the behavior was measured through nervous function examination and TTC staining after focal cerebral ischemia. The pathologic change and ultrastructure observation were adopted as a auxiliary measure for evaluating the therapeutic effect. The change of Caspase-3 expression was detected by in-situ hybridization. Finally we could estimate the therapeutic effect and mechanism of baicalin in rats with focal cerebral ischemia accurately.[Results] Baicalin could reduce the areas of cerebral infarction, alleviate encephaledema, decrease the expression of Caspase-3 in rat’s hippocampus and cortex after focal cerebral ischemia.[Conclusions] Baicalin has neural protective effect. To adjust the expression of Caspase-3 may be one of the most important mechanism of baicalin’s neural protective effect.
