回神颗粒对实验性大鼠局灶性脑缺血缺血区神经细胞凋亡的影响

摘要:
[目的]观察回神颗粒对大鼠局灶性脑缺血后缺血区神经细胞凋亡及调控基因Bcl-2、Bax蛋白的影响。[方法]选用成年雄性Wistar大鼠,应用开颅法烧灼闭塞大脑中动脉,制作局灶性脑缺血损伤动物模型(MACO)。动物随机分为假手术组和缺血组,缺血组又分为模型组和回神颗粒组。大鼠脑缺血7d后取脑并制作冰冻切片,进行原位末端缺口平移标记法(TUNEL)染色和Bax、Bcl-2免疫组织化学染色。[结果]治疗组大鼠脑组织TUNEL、Bax阳性细胞数明显较少,与模型组比存在显著性差异(P<0.01);治疗组大鼠脑组织Bcl-2阳性细胞数明显较多,与模型组比存在显著性差异(P<0.01);治疗组大鼠脑组织Bcl-2/Bax明显升高,与模型组比存在显著性差异(P<0.05)。[结论]回神颗粒具有抑制缺血性脑损伤后神经细胞凋亡的作用;回神颗粒提高MACO模型大鼠Bcl-2表达,抑制Bax表达,提高Bcl-2/Bax比值,可能是其抑制细胞凋亡,保护脑细胞的机制之一。

Effect of Hui-shen Granule on Apoptosis of Nerve Cells in Ischemic Region After Local Cerebral Ischemia in Rats

Abstract:
[Objective] To observe the effect of Hui-shen(restoring the mental function) capsule on apoptosis, Bcl-2 and Bax protein expression of nerve cells in brain after cerebral ischemia in rats. [Methods] We used Cauterization method to reproduce the MCAO model in Wistar rats and then divided the rats into 3 groups, 10 rats in each group: sham, Chinese herb of Hui-shen granule and control group. Frozen sections of the brain tissue were prepared in immunohistochemistry studies to investigate the expression of Bcl-2 and Bax protein and also in the TUNEL to observe the apoptosis of neural cells. [Results] There was a significant difference between the Hui-shen granule group and the model group with fewer TUNEL, Bax positive cells in the Hui-shen granule group(P<0.01). There was a significant difference between the Hui-shen granule group and the model group with more Bcl-2 positive cells in the Hui-shen granule group as well(P<0.01). there was also a significant difference between the Hui-shen granule group and the model group with higher level Bcl-2/Bax in the Hui-shen granule group(P<0.05). [Conclusions] Hui-shen granule can suppress apoptosis of cerebral ischemia-traumatic nervous cells; Hui-shen granule can enhance the expression of Bcl-2, suppress the expression of Bax and improve the ratio of Bcl-2 to Bax in the brain of rat with MCAO. This may be one of the mechanisms for Hui-shen granule in suppressing apoptosis and protecting the cell of brain.
