[目的]探索逆针灸关元穴对去卵巢大鼠下丘脑功能的调节机制。[方法]将3。5月龄SD雌性大鼠30只,随机分为正常组、假手术组、去卵巢模型组、逆灸组、逆针组。逆灸组、逆针组每周灸或针关元穴2次,共4周,4。5月龄时与去卵巢模型组同时行卵巢切除术,假手术组仅切除卵巢周围少量脂肪。采用免疫组化和原位杂交方法,观察各组大鼠下丘脑弓状核、室旁核、视上核热休克蛋白70(HSP70)和基因表达水平的变化。[结果]与正常组比较,下丘脑弓状核、室旁核、视上核HSP70及HSP70mRNA表达均升高;与假手术组比较,去卵巢模型组、逆灸组、逆针组HSP70及HSP70mRNA表达均升高;与去卵巢模型组比较,逆灸组、逆针组弓状核HSP70mRNA、室旁核、视上核HSP70及HSP70mRNA表达呈下降趋势,其中逆灸组、逆针组弓状核HSP70mRNA显著下降(P<0。01)。[结论]逆针灸关元穴能够影响去卵巢大鼠下丘脑不同核团HSP70及HSP70mRNA表达水平的变化,可能是其调节更年期机体神经内分泌紊乱的途径之一。 |

Effect of preventive acupuncture and preventive moxibustion at Guanyuan point on the expression of HSP70 and HSP70 mRNA in hypothalamus of ovariectomy rats |

[Objection] To investigate the influence of preventive acupuncture and preventive moxibustion at Guanyuan(CV4) point on the function of hypothalamus in ovariectomy rats.[Methods] Thirty female SD rats,aged 3.5 months,were randomly divided into normal group,sham operation group,ovariectomy model group,preventive acupuncture group and preventive moxibustion group.In preventive acupuncture and preventive moxibustion groups,Guanyuan was punctured with the needle retained for 20 min,and given with moxibustion for one moxa-cone respectively from aged 3.5th month.The treatment was conducted twice every week,4 weeks altogether.At 4.5th month,ovariectomy was made in the preventive acupuncture group,the preventive moxibustion group and the ovariectomy model group.For the sham operation group,only a little fat around the ovary was removed without ovariectomy,and nothing was not applied to the normal group.Immunohistochemical and in situs hybridization methods were used to investigate changes of expressions of HSP70 and HSP70 mRNA in arcuate nucleus,paraventricular nucleus and supraoptic nucleus.[Results] Compared to the normal group and sham operation group,the expressions of HSP70 and HSP70 mRNA in arcuate nucleus,paraventricular nucleus and supraoptic nucleus of hypothalamus increased in the ovariectomy model group,preventive acupuncture group and preventive moxibustion group.After preconditioning with acupuncture or moxibustion,the expression of HSP70 and HSP70 mRNA decreased,the expression of HSP70 mRNA in arcuate nucleus significantly decreased in preventive acupuncture group and preventive moxibustion group(P<0.01).[Conclusion] Preventive acupuncture and moxibustion at Guanyuan point have an effect on the expressions of HSP70 and HSP70 mRNAin different hypothalamic nuclei in ovariectomy rats,which possibly is one way for modulating the menopausal dysfunction. |
