消岩汤剂拆方配伍对MCF-7乳腺癌细胞生长及凋亡机制研究

摘要:

[目的]观察消岩汤和各拆方组药物对MCF-7乳腺癌细胞增殖活性,促进凋亡的作用,与化疗药物5-氟尿嘧啶(5-Fu)联合应用的效果。[方法]消岩汤全方和根据功效拆分的清热解毒组、活血化瘀组、解毒祛瘀组(清热解毒组+活血化瘀组)、益气扶正组和全方组。以噻唑蓝法(MTT)检测MCF-7乳腺癌细胞的细胞增殖活性,与5-Fu化疗药物联合作用的效果,用Hoechst33258法检测细胞的凋亡情况。[结果]各组均有不同程度地抑制细胞活性和促进凋亡,清热解毒组优于全方组优于解毒祛瘀组优于活血化瘀组优于益气扶正组,与5-Fu化疗药物合用表现出协同和相加作用。[结论]消岩汤在体外实验中能够抑制MCF-7乳腺癌细胞生长,促进凋亡,与5-Fu化疗药物合用有一定的化疗增效作用,清热解毒组药物在抑瘤和增效作用方面,与全方接近,提示清热解毒组中药为体外实验中全方抗肿瘤作用及化疗增效作用的有效组分。

Abstract:

[Objective] To investigate the growth and apoptosis of MCF-7 breast cancer cell treated by Xiaoyantang(Eliminating Cancer Decoction) and every group of dismantle prescription and the effect combined with 5-fluorouracil.[Methods] Based on the effect of the Eliminate Cancer Decoction it was the divided into five groups: clearing away heat and detoxication group,activating blood circulation and removing stasis group,detoxicationa removing stasis group(clearing heat combined with activating blood circulation and removing stasis),benefiting vital QI and strengthening the body resistance group and whole prescription group.To investigate the growth and apop-tosis of MCF-7 breast cancer the MTT was used and for detecting the apoptosis by eliminating cancer decoction combined with 5-fluo-rouracil Hoechst 33258 method was used.[Result] All the medicine of each group had effect to inhibit the cancer cell proliferation and to promote apoptosis in different degree.The effect of clearing away heat and detoxication group was superior to the whole decoction group.After that they were detoxication and removing stasis group,activating blood circulationa and removing stasis group,benefiting vital QI and strengthening body resistance.When using with 5-fluorouracil synergetic or additive effect could be found.[Conclusion] It is demon-strated in vitro experiment that the cancer eliminating decoction can suppress the MCF-7 breast cancer growth and promote the apoptosis.Using with 5-Fu chemotherapy together it has effect-increasing action.The inhibitory effect on growth or effect-increasing action of Clear-ing away heat and detoxication group was similar to the whole prescription group,suggesting that the medicine of clearing away heat and detoxication was the effective component of the whole prescription for anti-cancer and chemotherapy strengthening effect.
