维胃方对胃溃疡大鼠胃黏膜MPO、H+-K+-ATP酶活性的影响

摘要:
[目的]观察维胃方对胃溃疡大鼠胃黏膜髓过氧化物酶(MPO)和H+-K+-ATP酶的影响,探讨维胃方治疗胃黏膜损伤的可能机制。[方法]采用乙酸烧灼法,建立胃溃疡模型,用比色法测定胃黏膜MPO和H+-K+-ATP酶活性,并观察胃黏膜组织病理学变化。[结果]从胃黏膜大体标本、切片观察,维胃方中剂量组疗效最佳。与自愈组和模型组比较,维胃方各组胃黏膜MPO活性显著降低(P<0.01或P<0.05),其中以维胃方中剂量组值最小。维胃方各剂量组的H+-K+-ATP酶活性均高于自愈组(P<0.01或P<0.05),接近正常组(P>0.05),表明维胃方能恢复其活性。[结论]维胃方对大鼠实验性胃溃疡具有明显的保护作用,其机制可能是通过降低大鼠胃黏膜MPO活性,进而减轻炎症介质对胃黏膜的损伤。同时其能修复已损伤的胃黏膜,故能恢复H+-K+-ATP酶活性。

Effect of Weiwei prescription on activity of MPO and H+-K+-ATPase of gastric mucosa in rats with experimental gastric ulcer

Abstract:
[Objective] To investigate the effect of Weiweifang(WWF) on the activity of MPO and H+-K+-ATPase of gastric mucosa in rat with experimental gastric ulcer and to explore the possible mechanism of WWF in repairing stomach mucosa damage.[Methods] The model was induced by acetic acid and colorimetric method was used to determine the activity of MPO and H+-K+-ATPase in gastric mu-cosa.Meantime,the pathologic change of gastric mocosa was examined.[Results] The therapeutic effect of middle dose WWF group was the best on observing the gastric mucosa and tissue slice.Comparison of results among self-cure group,model group and experimental group found that WWF could inhibit MPO activity significantly(P<0.01 or P<0.05),especially in mid-dose group.The activity of H+-K+-ATPase could be raised by WWF because its activity were higher in cured groups of WWF than those in self-cure group(P<0.01 or P< 0.05) and similar to that of the normal group.[Conclusion] WWF could exert a significant protective effect against experimental gastric ulcer,possibly by decreasing MPO activity and ameliorating the gastric mucosa damage of inflammatory mediators,meanwhile,it could recover H+-K+-ATPase activity by repairing mucosa damage.
