丹酚酸B对猪冠状动脉收缩的影响

摘要:
[目的]观察丹酚酸B(SalB)对猪冠状动脉收缩的影响,并初步探讨其作用机制。[方法]采用离体平滑肌实验方法,观察丹酚酸B对氯化钾(KCl)、氯化钙(CaCl2)和组胺(Hist)诱导离体猪冠状动脉环收缩的影响,以及在无鲈离子(Ca2+)克氏液中,对组胺引起猪冠状动脉环第一相收缩和Ca2+引起第二相收缩的影响。[结果]丹酚酸B可使KCl、CaCl2和Hist诱导猪冠脉环收缩的量-效曲线非平行右移,最大效应降低,呈非竞争性拮抗作用;对冠脉环第一相和第二相收缩均有明显的抑制作用。[结论]丹酚酸B具有扩张冠状动脉的作用,其机制可能与抑制血管平滑肌细胞外钙内流和内钙释放有关。

Abstract:
[Objective] To observe the effect of salvianolic acid B(Sal B) on the contraction of porcine coronary artery strips,and investi-gate its mechanism of action.[Methods] Using experimental method of smooth muscle in vitro the effect of Sal B on the contraction of porcine coronary artery strips induced by KCL,CaCl2 and Hist was observed.[Results] Salvianolic acid B had nonparallel right migration effect on KCl,CaCl2 and Histamine induced dose-response curves,and decrease the maximal effect.Moreover,it can greatly inhibit in-tracellular Ca2+-dependent contraction induced by Histamine,and show relatively little effect on the extracellular Ca2+-dependent contrac-tion induced by CaCl2.[Conclusion] Salvianolic acid B had a significant relaxing effect on contraction of porcine coronary artery,and had an antagonistic effect on the two kinds of contractions.
