[目的]考察银杏内酯B(GB)通过作用于脑微环境内星形胶质细胞(AC)对神经干细胞(NSCs)趋化的影响,并探讨其作用机制。[方法]利用Millicell小室建立体外趋化迁移模型,采用糖氧剥夺(OGD)方法损伤AC。GB(10、20、40μM)作用于AC 12 h,计数趋化迁移至Millicell小室下层细胞数目;采用反转录聚合酶链反应(RT-PCR)检测GB对AC趋化相关基因VEGF mRNA表达的影响。[结果]GB(10、20μM)作用于AC可显著增加GFP-NSC定向迁移数目(P<0.05)。RT-PCR结果显示,GB(102、0μM)可显著上调AC VEGF mRNA表达(P<0.05)。[结论]GB可以促进NSCs向损伤的AC趋化,这一过程可能是由VEGF介导的。 |

In vitro study on effects of astrocytes following treatment with ginkgolide B on chemotaxis of neural stem cells |

Abstract: |
[Objective] To study the effect of ginkgolide B(GB) on chemotaxis of neurall stem cells(NSCs) Through acting on the astrocyte in brain microcirculation and its mechanism.[Methods] Using a millicell Chamber the chemotactic migration was observed.Oxygen-glu-cose deprivation(GOD) was used for making the astrocyte cell injured.10,20 μM of GB were treated on AC for 12 hrs and then the num-ber migrated on the lower layer of Millicell chmber was counted.The expression of chemotaxis gene VEGF mRNA of AC was detected by reverse transcription polymerase chain reaction(RT-PCR).[Results] The migrated number of the GFP-NSC after acting on AC with GB(10,20 μM)were significantly increased(P<0.05).And the results of RT-PCR showed that the VEGF mRNA expression were up-regulated significantly by GB(10,20 μM).[Concluision] GB can promote the NSCs chemokinesis and increase migrating to the injuried AC.This process may be introduced by VEGF. |
