五田保肝液对酒精性肝纤维化大鼠肝组织TGF-β1表达影响的实验研究

摘要:

[目的]五田保肝液对酒精性肝纤维化大鼠肝组织转化生长因子β1(TGF-β1)表达的影响。[方法]用白酒辅以吡唑、玉米油的混合食料灌胃的方法建立大鼠酒精性肝纤维化模型,随机分为正常组、模型组、中药低剂量组、中剂量组、高剂量组、易善复组(正常组10只,其余组各18只)。12周后处死苏木精-伊红(HE)、Masson染色观察肝纤维化的形成;酶联免疫检测法(ELISA)检测血清Ⅲ型前胶原蛋白肽(PⅢP)、层黏连蛋白(LN)两者含量;实时定量逆转录-聚合酶链反应(RT-PCR)检测肝组织中TGF-β1 mRNA的表达。[结果]模型组大鼠血清Ⅲ型前胶原蛋白肽(PⅢP)、层黏连蛋白(LN)含量和肝组织TGF-β1mRNA的表达明显增多,各治疗组相对于模型组均有不同程度减少,其中五田保肝液中、高剂量组降低更为明显。[结论]五田保肝液可抑制酒精性肝纤维化大鼠肝组织TGF-β1的表达,这可能是其防治酒精性肝纤维化的机制之一。

Abstract:

[Objective] To observe the effect of Wutian Baogan solution on the expression of liver tissue TGF-β1 in rats with alcoholic liver fibrosis.[Methods] The rat model of alcoholic liver fibrosis was established by gastric infusion of alcohol supplemented with pyrazole,corn oil.The Wistar rats with hepatic fibrosis were randomly divided into normal group,model group,low-dose group,middle dose group,high-dose group,Essentiale group.After treating for 12 weeks the rats were killed to stain the hepatic tissue with HE and Masson.Using enzyme-linked immunosorbent assay(ELISA) the content of typeⅢ collagen precursor protein(PⅢP) and laminin(LN) was determined.With Real-time RT-PCR the liver tissue transforming growth factor β1(TGF-β1) mRNA expression was detected.[Results] The serum PⅢP,LN and liver TGF-β1mRNA expression of model group was significantly increased,relative decreasing was found in each treatment group when compared with model group.Among the treatment group with Wutian Baogan solution the middle or high dosage group had more obvious decreasing.[Conclusion] Wutian Baogan solution can inhibit the expression of liver tissue TGF-β1 in rats with alcoholic liver fibrosis.It may be one of the mechanisms of prevention and treatment of alcoholic liver fibrosis.
