[目的]观察早期应用中药复方对哮喘气道炎症和气道重塑的治疗作用。[方法]40只SD大鼠随机分为4组:正常组、哮喘组、布地奈德(BUD)组及止喘汤组,采用卵蛋白(OVA)致敏加激发法复制慢性哮喘模型,观察各组大鼠外周血(PB)、支气管肺泡灌洗液(BAL)中嗜酸性粒细胞(EOS)计数的变化情况,同时对肺组织切片行苏木精-伊红(HE)染色测定支气管壁周围EOS个数及气道壁厚度。[结果]哮喘组与正常组比较:PB、BAL和气道壁周围EOS的计数水平明显升高,气道壁明显增厚(P<0.01)。药物干预后,干预组与哮喘组比较:PB、BAL和气道壁周围EOS的计数水平明显降低,气道壁明显变薄(P<0.01);且药物干预组之间相比较无统计学差异(P>0.05)。[结论]止喘汤不但能抑制哮喘气道炎症,而且还具有干预气道重塑的作用。 |

Effect of Zhichuan decoction on airway inflammation and remodeling in asthmatic rats |

Abstract: |
[Objective] To investigate the effects of early treatment with Zhichuan decoction on airway inflammation and remodeling in asthmatic rats.[Methods] Forty Sprague-Dawley rats were randomly divided into four groups equally:normal control group,asthmatic group,budesonide aerosol group and Zhichuan decoction group.The model of asthma was established by OVA(ovalbumin) sensitizing and challenging.The differential counts of inflammatory cell in peripheral blood(PB) and bronchoaveolar(BAL) portion were calculated.By hematoxylin and eosin(HE) stain,pathologic changes of lung tissue were observed,the counts of eosinophil(EOS) and the airway thickness were measured.[Results] In asthmatic group,EOS counts in PB,BAL,airway around and the airway thickness were increased significantly when compared with normal control group(P<0.01).After intervention with budesonide aerosol and Zhichuan decoction,the above indicators were significantly decreased when compared with asthmatic models(P<0.01);No significant difference was found between the two treatment groups(P>0.05).[Conclusion] Prolonged allergen exposure can induce chronic asthma,intervention with Zhichuan decoction can inhibit the airway inflammation and prevent airway remodeling. |
