[目的]探讨中药丹酚酸B预处理的内皮祖细胞(EPCs)对急性心肌梗死(AMI)大鼠移植骨髓间充质干细胞(BMSCs)后心肌微环境的影响。[方法]56只AM I模型大鼠随机分为假手术组、模型组、BM SCs组、EPCs+BM SCs组,其中治疗组用丹酚酸B最佳药物浓度处理的EPCs与BM SCs混合,在大鼠AM I区周边分5点注射。免疫荧光和免疫组化方法检测AM I大鼠梗死区心肌收缩蛋白α型肌球蛋白重链(α-MHC)和I型胶原蛋白的表达。[结果]细胞移植4周后,各细胞联合移植组中,2:14、:1和8:1组在Brdu集中表达区域均有不同强弱程度的α-MHC阳性表达,除假手术组较少外,所有接受冠状动脉结扎的大鼠心肌梗死区I型胶原表达均升高。[结论]BMSCs在丹酚酸B预处理的EPCs干预的心肌微环境下可向心肌细胞分化,可预防或改善心室重塑的发生。 |

[Objective] To investigate the effect of salvianolic acid B preconditioned endothelial progenitor cells(EPCs) combined with implantation of bone mesenchymal stem cells(BMSCs) on the gene expression in rats with acute myocardial infarction(AMI).[Methods] Forty-two AMI rats were divided randomly into sham operation model,BMSCs and EPCs combined with BMSCs transplantation group,and EPCs preconditioned with salvianolic acid B combined with transplantation of BMSCs were injected into five points of heart ischemia area.[Result] After 4 weeks,among each combined cell transplantation group,in 2∶1,4∶1 and 8∶1 group in the area of Brdu concentrated expression positive α-MHC expression at different degree could be found.Except that the sham operation group had less expression,all the area with myocardial infarction underwent coronary artery ligation in rats the type I collagen expression increased.[Conclusion] Salvianolic acid B-preconditioned EPCs with transplanted BMSCs under the microenvironment of myocardial muscle can be differentiated into myocardial cells.They can prevent or improve the occurrence of ventricular remodeling. |
