补中益气汤对甲减肾损害模型大鼠肾脏功能、形态及VEGF表达的影响

摘要:
[目的]拟观察甲状腺功能减退时肾脏功能、形态及血管内皮细胞生长因子(VEGF)表达变化及补中益气汤对其的影响。[方法]将大鼠按随机数字表法随机分组,56 d后测定血清甲状腺功能[总三碘甲状腺原氨酸(TT3)、总甲状腺素(TT4)及促甲状腺激素(TSH)]、血清肌酐(SCr)、尿素氮(月哉晕)水平;光镜下观察肾脏形态,以免疫组化检测VEGF表达水平。[结果]1)与正常对照组相比左旋甲状腺素(L-T4)组及模型组血清BUN均增高(P<0.05),SCr也均增高(P<0.05)。圆)肾脏VEGF光密度:与正常对照组相比各组均无明显差异,与模型组相比只有补中益气汤组光密度明显升高(孕约园援园员)。3)PAS染色比较:处理21 d时,模型组呈轻度系膜增生,用药56 d后模型组有中度增生,有部分毛细血管扩张,两用药组以补中益气组增生最轻,L-T4组增生较模型组明显减轻,仍可见毛细血管扩张。[结论]补中益气汤能够促进VEGF分泌,VEGF升高对甲减肾损害具有保护作用。

Effect of Buzhong Yiqi decoction on the kidney function,morphology and VEGF expression in rats with hypothyroid kidney damages

Abstract:
[Objective] To study effect of Buzhong Yiqi decoction on the kidney function,morphology and VEGF expression in rats with hypothyroid kidney damages.[Methods] The rats were divided randomly into experiment group and normal group.56 days later we detected the level of serum TT3,TT4,TSH,SCr and BUN,observed the morphologic change of the kidney and the VEGF expression.[Results] 1)Compared with normal control group,the level of serum BUN(P<0.05) and SCr(P<0.05) in L-T4 group and model increased.2)About VEGF optical density,compared with normal control group,each group had no obvious difference.Compared with model group,Buzhong Yiqi decoction group increased obviously(P<0.01).3)About PAS staining,on the 21 days of treatment,model group displayed slightly hyperplasia;after 56 days treatment,model group displayed medium hyperplasia,the hyperplasia in Buzhong Yiqi decoction group was most slight among two treatment groups.[Conclusion] The conceivable mechanism of hypothyroid kidney damages is the loss of the glomerular capillaries.Buzhong Yiqi decoction can increase the endocrine function of VEGF and the rising VEGF in kidney plays a protective role for hypothyroidisn.
