白附子混悬液对荷瘤小鼠p16、bcl-2的表达及端粒酶活性的相关性研究

摘要:
[目的]探讨白附子混悬液(SGTR)对荷瘤小鼠端粒酶活性的调节及对p16、bcl-2的相关表达的研究。[方法]采用酶联免疫吸附实验(ELISA)法检测小鼠血清中端粒酶浓度;用免疫组织化学方法测定H22肿瘤组织中p16和bcl-2蛋白的表达情况来推断SGTR对肿瘤细胞凋亡机制的作用。[结果]SGTR可上调p16蛋白表达水平,差异有统计学意义(P<0.01);下调bcl-2蛋白的表达水平(P<0.01),抑制H22荷瘤小鼠体内端粒酶活性(P<0.01)。[结论]SGTR通过抑制端粒酶活性、下调凋亡基因bcl-2的表达及上调抑癌基因p16的表达从而促进肿瘤细胞凋亡来达到抗肿瘤的作用。

Study on correlation between suspension of white monkshood(SGTR) stimulated expression of p16,bcl-2 and telomerase activity in tumor-bearing mice

Abstract:
[Objective] To explore the effect of suspension of white monkshood(SGTR) on telomerase activity and p16,bcl-2 expression in tumor-bearing mice.[Methods] The telomerase concentration in mice serum was detected with ELISA method.The expression of p16 and bcl-2 protein in H22 tumor tissues was determined with immunohistochemistry method and then the effect mechanism of white monkshood on the tumor apoptosis was inferred.[Results] The expression level of p16 gene could be up-regulated by SGTR and it had significantly difference when compared with model group(P<0.01);the expression level of p16 gene could be down-regulated by SGTR and it had significantly difference when compared with model group(P<0.01);the telomerase activity in tumor-bearing mice was inhibited by SGTR and it had significantly difference when compared with model group(P<0.01).[Conclusion] SGTR can performed its anti-tumor effect by inhibiting the telomerase activity,down-regulating the expression of apoptosis gene bcl-2,up-regulating the cancer suppressor gene p-16 and thereby promoting the apoptosis of tumor cells.
